This assay has high sensitivity and excellent specificity for detection of Human WFDC2. No significant cross-reactivity or interference between Human WFDC2 and analogues was observed.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with Avidin. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody and Horseradish Peroxidase (HRP) -conjugated antibody preparation specific for HE4 and incubated. Then substrate solutions are added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of HE4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a protein that is a member of the WFDC domain family. The WFDC domain, or WAP Signature motif, contains eight cysteines forming four disulfide bonds at the core of the protein, and functions as a protease inhibitor in many family members. This gene is expressed in pulmonary epithelial cells, and was also found to be expressed in some ovarian cancers. The encoded protein is a small secretory protein, which may be involved in sperm maturation. [provided by RefSeq, Jul 2008]
