Principle of the assay: </b<This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CCL5 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CCL5 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CCL5 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CCL5 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The superfamily is divided into four subfamilies based on the arrangement of the N-terminal cysteine residues of the mature peptide. This chemokine, a member of the CC subfamily, functions as a chemoattractant for blood monocytes, memory T helper cells and eosinophils. It causes the release of histamine from basophils and activates eosinophils. This cytokine is one of the major HIV-suppressive factors produced by CD8+ cells. It functions as one of the natural ligands for the chemokine receptor chemokine (C-C motif) receptor 5 (CCR5), and it suppresses in vitro replication of the R5 strains of HIV-1, which use CCR5 as a coreceptor.
