Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CA3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CA3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CA3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CA3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Carbonic anhydrase III (CAIII) is a member of a multigene family (at least six separate genes are known) that encodes carbonic anhydrase isozymes. These carbonic anhydrases are a class of metalloenzymes that catalyze the reversible hydration of carbon dioxide and are differentially expressed in a number of cell types. The expression of the CA3 gene is strictly tissue specific and present at high levels in skeletal muscle and much lower levels in cardiac and smooth muscle. A proportion of carriers of Duchenne muscle dystrophy have a higher CA3 level than normal. The gene spans 10.3 kb and contains seven exons and six introns. [provided by RefSeq, Oct 2008]
