Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for beta-APP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any beta-APP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for beta-APP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of beta-APP bound in the initial step. The color development is stopped and the intensity of the color is measured.

Some of these peptides are secreted and can bind to the acetyltransferase complex APBB1/TIP60 to promote transcriptional activation, while others form the protein basis of the amyloid plaques found in the brains of patients with Alzheimer disease. In addition, two of the peptides are antimicrobial peptides, having been shown to have bacteriocidal and antifungal activities. Mutations in this gene have been implicated in autosomal dominant Alzheimer disease and cerebroarterial amyloidosis (cerebral amyloid angiopathy).
