Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HMG-CoAR has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HMG-CoAR present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HMG-CoAR is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HMG-CoAR bound in the initial step. The color development is stopped and the intensity of the color is measured.

HMG-CoA reductase is the rate-limiting enzyme for cholesterol synthesis and is regulated via a negative feedback mechanism mediated by sterols and non-sterol metabolites derived from mevalonate, the product of the reaction catalyzed by reductase. Normally in mammalian cells this enzyme is suppressed by cholesterol derived from the internalization and degradation of low density lipoprotein (LDL) via the LDL receptor. Competitive inhibitors of the reductase induce the expression of LDL receptors in the liver, which in turn increases the catabolism of plasma LDL and lowers the plasma concentration of cholesterol, an important determinant of atherosclerosis.
