Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MAGE-3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MAGE-3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MAGE-3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MAGE-3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the MAGEA gene family. The members of this family encode proteins with 50 to 80% sequence identity to each other. The promoters and first exons of the MAGEA genes show considerable variability, suggesting that the existence of this gene family enables the same function to be expressed under different transcriptional controls. The MAGEA genes are clustered at chromosomal location Xq28. They have been implicated in some hereditary disorders, such as dyskeratosis congenita. [provided by RefSeq, Jul 2008]
