Priciple of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for sLOX-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sLOX-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for sLOX-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sLOX-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Receptor that mediates the recognition, internalization and degradation of oxidatively modified low density lipoprotein (oxLDL) by vascular endothelial cells. OxLDL is a marker of atherosclerosis that induces vascular endothelial cell activation and dysfunction, resulting in pro-inflammatory responses, pro-oxidative conditions and apoptosis. Its association with oxLDL induces the activation of NF-kappa-B through an increased production of intracellular reactive oxygen and a variety of pro-atherogenic cellular responses including a reduction of nitric oxide (NO) release, monocyte adhesion and apoptosis. In addition to binding oxLDL, it acts as a receptor for the HSP70 protein involved in antigen cross-presentation to naive T-cells in dendritic cells, thereby participating in cell-mediated antigen cross-presentation.
