Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for PON3 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any PON3 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for PON3 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of PON3 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the paraoxonase family and lies in a cluster on chromosome 7 with the other two family members. The encoded protein is secreted into the bloodstream and associates with high-density lipoprotein (HDL). The protein also rapidly hydrolyzes lactones and can inhibit the oxidation of low-density lipoprotein (LDL), a function that is believed to slow the initiation and progression of atherosclerosis. Alternatively spliced variants which encode different protein isoforms have been described; however, only one has been fully characterized. [provided by RefSeq, Jul 2008]
