For Sample: For the quantitative determination of human NAD (P) H dehydrogenase, quinone 1 (NQO1) concentrations in serum, plasma, tissue homogenates. PRINCIPLE OF THE ASSAY This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for NQO1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any NQO1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for NQO1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of NQO1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a member of the NAD(P)H dehydrogenase (quinone) family and encodes a cytoplasmic 2-electron reductase. This FAD-binding protein forms homodimers and reduces quinones to hydroquinones. This protein's enzymatic activity prevents the one electron reduction of quinones that results in the production of radical species. Mutations in this gene have been associated with tardive dyskinesia (TD), an increased risk of hematotoxicity after exposure to benzene, and susceptibility to various forms of cancer. Altered expression of this protein has been seen in many tumors and is also associated with Alzheimer's disease (AD).
