Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for G6PD has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any G6PD present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for G6PD is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of G6PD bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes glucose-6-phosphate dehydrogenase. This protein is a cytosolic enzyme encoded by a housekeeping X-linked gene whose main function is to produce NADPH, a key electron donor in the defense against oxidizing agents and in reductive biosynthetic reactions. G6PD is remarkable for its genetic diversity. Many variants of G6PD, mostly produced from missense mutations, have been described with wide ranging levels of enzyme activity and associated clinical symptoms. G6PD deficiency may cause neonatal jaundice, acute hemolysis, or severe chronic non-spherocytic hemolytic anemia. Two transcript variants encoding different isoforms have been found for this gene.
