products description :
Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to CTSS. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for CTSS and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain CTSS, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of CTSS in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
The protein encoded by this gene, a member of the peptidase C1 family, is a lysosomal cysteine proteinase that may participate in the degradation of antigenic proteins to peptides for presentation on MHC class II molecules. The encoded protein can function as an elastase over a broad pH range in alveolar macrophages. Alternatively spliced transcript variants encoding distinct isoforms have been found for this gene. [provided by RefSeq, Dec 2010]
uniprot summary :
Function: Thiol protease. Key protease responsible for the removal of the invariant chain from MHC class II molecules. The bond-specificity of this proteinase is in part similar to the specificities of cathepsin L and cathepsin N. Catalytic activity: Similar to cathepsin L, but with much less activity on Z-Phe-Arg- -NHMec, and more activity on the Z-Val-Val-Arg- -Xaa compound.
