This assay has high sensitivity and excellent specificity for detection of Rat PRG4. No significant cross-reactivity or interference between Rat PRG4 and analogues was observed.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to PRG4. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for PRG4 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain PRG4, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of PRG4 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

The protein encoded by this gene is a large proteoglycan specifically synthesized by chondrocytes located at the surface of articular cartilage, and also by some synovial lining cells. This protein contains both chondroitin sulfate and keratan sulfate glycosaminoglycans. It functions as a boundary lubricant at the cartilage surface and contributes to the elastic absorption and energy dissipation of synovial fluid. Mutations in this gene result in camptodactyly-arthropathy-coxa vara-pericarditis syndrome. Multiple transcript variants encoding different isoforms have been found for this gene. Function: Plays a role in boundary lubrication within articulating joints.
