This assay has high sensitivity and excellent specificity for detection of Human COLEC11. No significant cross-reactivity or interference between Human COLEC11 and analogues was observed.

Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to COLEC11. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for COLEC11 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain COLEC11, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of COLEC11 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

This gene encodes a member of the collectin family of C-type lectins that possess collagen-like sequences and carbohydrate recognition domains. Collectins are secreted proteins that play important roles in the innate immune system by binding to carbohydrate antigens on microorganisms, facilitating their recognition and removal. The encoded protein binds to multiple sugars with a preference for fucose and mannose. Mutations in this gene are a cause of 3MC syndrome-2. Function: Lectin that binds to various sugars including fucose and mannose. Has a higher affinity for fucose compared to mannose. Does not bind to glucose, N-acetylglucosamine and N-acetylgalactosamine. Also binds lipopolysaccharides (LPS).
