Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MUC2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MUC2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MUC2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MUC2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the mucin protein family. Mucins are high molecular weight glycoproteins produced by many epithelial tissues. The protein encoded by this gene is secreted and forms an insoluble mucous barrier that protects the gut lumen. The protein polymerizes into a gel of which 80% is composed of oligosaccharide side chains by weight. The protein features a central domain containing tandem repeats rich in threonine and proline that varies between 50 and 115 copies in different individuals.
