Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MMP-13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MMP-13 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MMP-13 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MMP-13 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a member of the matrix metalloproteinase family that plays a role in wound healing, skeletal development and bone remodeling. The encoded protein is activated by the removal of an N-terminal activation peptide to generate a zinc-dependent endopeptidase enzyme that can cleave various native collagens, including types I - IV, X and XIV. Mice lacking the encoded protein display profound defects in growth plate cartilage as well as a delay in the endochondral bone development. Lack of the encoded protein also impairs the wound healing process due to reduced keratinocyte migration and vascular density at the wound site.
