This assay has high sensitivity and excellent specificity for detection of guinea pig IL-13. No significant cross-reactivity or interference between guinea pig IL-13 and analogues was observed.

Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 7. 5 pg/ml -300 pg/ml. Sensitivity: The minimum detectable dose of guinea pig IL-13 is typically less than 3.75 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest guinea pig IL-13 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<15%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay: Inter-assay Precision (Precision between assays): CV%<15%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for IL-13 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells with a Horseradish Peroxidase (HRP) conjugated antibody specific for IL-13. Following a wash to remove any unbound reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-13 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes an immunoregulatory cytokine produced primarily by activated Th2 cells. This cytokine is involved in several stages of B-cell maturation and differentiation. It up-regulates CD23 and MHC class II expression, and promotes IgE isotype switching of B cells. This cytokine down-regulates macrophage activity, thereby inhibits the production of pro-inflammatory cytokines and chemokines. This cytokine is found to be critical to the pathogenesis of allergen-induced asthma but operates through mechanisms independent of IgE and eosinophils. This gene, IL3, IL5, IL4, and CSF2 form a cytokine gene cluster on chromosome 5q, with this gene particularly close to IL4. [provided by RefSeq, Jul 2008]
