Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with antigen. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated anti-TPO antibody. A competitive inhibition reaction is launched between anti-TPO antibody (Standards or samples) and Biotin-conjugated anti-TPO antibody with the pre-coated antigen. The more amount of anti-TPO antibody in samples, the less pre-coated antigen bound by Biotin-conjugated anti-TPO antibody. After washing, Horseradish Peroxidase (HRP)-conjugated avidin is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of anti-TPO antibody in the sample. The color development is stopped and the intensity of the color is measured.

This gene encodes a membrane-bound glycoprotein. The encoded protein acts as an enzyme and plays a central role in thyroid gland function. The protein functions in the iodination of tyrosine residues in thyroglobulin and phenoxy-ester formation between pairs of iodinated tyrosines to generate the thyroid hormones, thyroxine and triiodothyronine. Mutations in this gene are associated with several disorders of thyroid hormonogenesis, including congenital hypothyroidism, congenital goiter, and thyroid hormone organification defect IIA.
