Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HB-EGF has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HB-EGF present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HB-EGF is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HB-EGF bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Growth factor that mediates its effects via EGFR, ERBB2 and ERBB4. Required for normal cardiac valve formation and normal heart function. Promotes smooth muscle cell proliferation. May be involved in macrophage-mediated cellular proliferation. It is mitogenic for fibroblasts, but not endothelial cells. It is able to bind EGF receptor/EGFR with higher affinity than EGF itself and is a far more potent mitogen for smooth muscle cells than EGF. Also acts as a diphtheria toxin receptor.
