Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for OT/BGP has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any OT/BGP present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for OT/BGP is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of OT/BGP bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a highly abundant bone protein secreted by osteoblasts that regulates bone remodeling and energy metabolism. The encoded protein contains a Gla (gamma carboxyglutamate) domain, which functions in binding to calcium and hydroxyapatite, the mineral component of bone. Serum osteocalcin levels may be negatively correlated with metabolic syndrome. Read-through transcription exists between this gene and the neighboring upstream gene, PMF1 (polyamine-modulated factor 1), but the encoded protein only shows sequence identity with the upstream gene product.
