The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest human 6-K-PGF1a concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to 6-K-PGF1a. Standards or samples are added to the appropriate microtiter plate wells with Biotin-conjugated 6-K-PGF1a. A competitive inhibition reaction is launched between 6-K-PGF1a (Standards or samples) and Biotin-conjugated 6-K-PGF1a with the pre-coated antibody specific for 6-K-PGF1a. The more amount of 6-K-PGF1a in samples, the less antibody bound by Biotin-conjugated 6-K-PGF1a. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of 6-K-PGF1a in the sample. The color development is stopped and the intensity of the color is measured.
