Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TrpV1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TrpV1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TrpV1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TrpV1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Capsaicin, the main pungent ingredient in hot chili peppers, elicits a sensation of burning pain by selectively activating sensory neurons that convey information about noxious stimuli to the central nervous system. The protein encoded by this gene is a receptor for capsaicin and is a non-selective cation channel that is structurally related to members of the TRP family of ion channels. This receptor is also activated by increases in temperature in the noxious range, suggesting that it functions as a transducer of painful thermal stimuli in vivo.
