This assay has high sensitivity and excellent specificity for detection of human sCD14. No significant cross-reactivity or interference between human sCD14 and analogues was observed.

Samples: Serum, plasma, cell culture supernates, tissue homogenates, urine. Assay Type: Quantitative Sandwich. Sensitivity: Typically less than 0.229 ng/ml.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for sCD14 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any sCD14 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for sCD14 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of sCD14 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a surface antigen that is preferentially expressed on monocytes/macrophages. It cooperates with other proteins to mediate the innate immune response to bacterial lipopolysaccharide. Alternative splicing results in multiple transcript variants encoding the same protein. Function: In concert with LBP, binds to monomeric lipopolysaccharide and delivers it to the MD-2/TLR4 complex, thereby mediating the innate immune response to bacterial lipopolysaccharide (LPS). Acts via MyD88, TIRAP and TRAF6, leading to NF-kappa-B activation, cytokine secretion and the inflammatory response.
