specificity :
This assay has high sensitivity and excellent specificity for detection of human GCLC. No significant cross-reactivity or interference between human GCLC and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 0.156 ng/ml -10 ng/ml. Sensitivity: The minimum detectable dose of human GCLC is typically less than 0.039 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
. Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for GCLC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any GCLC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for GCLC is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of GCLC bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
Glutamate-cysteine ligase, also known as gamma-glutamylcysteine synthetase is the first rate-limiting enzyme of glutathione synthesis. The enzyme consists of two subunits, a heavy catalytic subunit and a light regulatory subunit. This locus encodes the catalytic subunit, while the regulatory subunit is derived from a different gene located on chromosome 1p22-p21. Mutations at this locus have been associated with hemolytic anemia due to deficiency of gamma-glutamylcysteine synthetase and susceptibility to myocardial infarction.[provided by RefSeq, Oct 2010]
