Principle of the Assay This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for ANX-I has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any ANX-I present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for ANX-I is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of ANX-I bound in the initial step. The color development is stopped and the intensity of the color is measured.

Annexin I belongs to a family of Ca(2+)-dependent phospholipid binding proteins which have a molecular weight of approximately 35,000 to 40,000 and are preferentially located on the cytosolic face of the plasma membrane. Annexin I protein has an apparent relative molecular mass of 40 kDa, with phospholipase A2 inhibitory activity. Since phospholipase A2 is required for the biosynthesis of the potent mediators of inflammation, prostaglandins and leukotrienes, annexin I may have potential anti-inflammatory activity.
