This assay has high sensitivity and excellent specificity for detection of Human MEPE. No significant cross-reactivity or interference between Human MEPE and analogues was observed.

Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MEPE has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MEPE present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MEPE is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MEPE bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a secreted calcium-binding phosphoprotein that belongs to the small integrin-binding ligand, N-linked glycoprotein (SIBLING) family of proteins. Members of this family are components of the extracellular matrix of bone and dentin and regulate bone mineralization. Deficiency of a similar protein in mouse results in increased bone mass.
