Introduction: Osteonectin is a glycoprotein in the bone that binds calcium. It is secreted by osteoblasts during bone formation, initiating mineralization and promoting mineral crystal formation. Osteonectin also shows affinity for collagen in addition to bone mineral calcium. Some current research shows a correlation between osteonectin over expression and ampullary cancers and chronic pancreatitis. Osteonectin is an acidic, secreted extracellular matrix glycoprotein that plays a vital role in bone mineralization, cell-matrix interactions, and collagen binding. Osteonectin also increases the production and activity of matrix metalloproteinases, a function important to invading cancer cells within bone. Additional functions of osteonectin beneficial to tumor cells include angiogenesis, proliferation and migration. Overexpression of osteonectin is reported in many Mouse cancers such as breast, prostate and colon. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to ON. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for ON and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain ON, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of ON in the samples is then determined by comparing the O.D. of the samples to the standard curve.
