Introduction: VEGF receptors are receptors for vascular endothelial growth factor (VEGF). VEGF Receptor 1 (also known as FLT) belongs to the src gene family and shows tyrosine protein kinase activity that is important for the control of cell proliferation and differentiation. The protein acts as a receptor for VEGF, VEGFB and PGF. An alternatively spliced form of the gene produces a soluble protein (sVEGFR-1/sFlt1) which binds VEGF with high affinity. sVEGFR-1 has a higher affinity for VEGF indicating that it may function as an inhibitor in the VEGF response. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to sVEGFR-1. Standards or samples are then added to the appropriate microtiter plate wells with a biotin-conjugated antibody preparation specific for sVEGFR-1 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5 tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain sVEGFR-1, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of sVEGFR-1 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

VEGFR family members are receptor tyrosine kinases (RTKs) which contain an extracellular ligand-binding region with seven immunoglobulin (Ig)-like domains, a transmembrane segment, and a tyrosine kinase (TK) domain within the cytoplasmic domain. This protein binds to VEGFR-A, VEGFR-B and placental growth factor and plays an important role in angiogenesis and vasculogenesis. Expression of this receptor is found in vascular endothelial cells, placental trophoblast cells and peripheral blood monocytes. Multiple transcript variants encoding different isoforms have been found for this gene. Isoforms include a full-length transmembrane receptor isoform and shortened, soluble isoforms.
