uniprot entry name :
GRAB_HUMAN
specificity :
This assay has high sensitivity and excellent specificity for detection of human Gzms-B. No significant cross-reactivity or interference between human Gzms-B and analogues was observed.
storage stability :
Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.
other info1 :
Samples: Serum, plasma, tissue homogenates. Assay Type: Sandwich. Detection Range: 1.56 pg/ml -100 pg/ml. Sensitivity: The minimum detectable dose of human Gzms-B is typically less than 0.39 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest protein concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.
other info2 :
Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<10%. Three samples of known concentration were tested in twenty assays to assess.
products description :
Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for Gzms-B has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Gzms-B present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Gzms-B is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Gzms-B bound in the initial step. The color development is stopped and the intensity of the color is measured.

ncbi summary :
Cytolytic T lymphocytes (CTL) and natural killer (NK) cells share the remarkable ability to recognize, bind, and lyse specific target cells. They are thought to protect their host by lysing cells bearing on their surface 'nonself' antigens, usually peptides or proteins resulting from infection by intracellular pathogens. The protein encoded by this gene is crucial for the rapid induction of target cell apoptosis by CTL in cell-mediated immune response. [provided by RefSeq, Jul 2008]
uniprot summary :
Function: This enzyme is necessary for target cell lysis in cell-mediated immune responses. It cleaves after Asp. Seems to be linked to an activation cascade of caspases (aspartate-specific cysteine proteases) responsible for apoptosis execution. Cleaves caspase-3, -7, -9 and 10 to give rise to active enzymes mediating apoptosis. Catalytic activity: Preferential cleavage: -Asp- -Xaa- >> -Asn- -Xaa- > -Met- -Xaa-, -Ser- -Xaa-.1 PublicationManual assertion based on experiment in:Ref.12. Enzyme regulation: Inactivated by the serine protease inhibitor diisopropylfluorophosphate.1 PublicationManual assertion based on experiment in:Ref.12
