Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for TGM2 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any TGM2 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for TGM2 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of TGM2 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Transglutaminases are enzymes that catalyze the crosslinking of proteins by epsilon-gamma glutamyl lysine isopeptide bonds. While the primary structure of transglutaminases is not conserved, they all have the same amino acid sequence at their active sites and their activity is calcium-dependent. The protein encoded by this gene acts as a monomer, is induced by retinoic acid, and appears to be involved in apoptosis. Finally, the encoded protein is the autoantigen implicated in celiac disease.
