Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for MPO has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any MPO present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for MPO is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of MPO bound in the initial step. The color development is stopped and the intensity of the color is measured.

Myeloperoxidase (MPO) is a heme protein synthesized during myeloid differentiation that constitutes the major component of neutrophil azurophilic granules. Produced as a single chain precursor, myeloperoxidase is subsequently cleaved into a light and heavy chain. The mature myeloperoxidase is a tetramer composed of 2 light chains and 2 heavy chains. This enzyme produces hypohalous acids central to the microbicidal activity of netrophils. [provided by RefSeq, Jul 2008]

Function: Part of the host defense system of polymorphonuclear leukocytes. It is responsible for microbicidal activity against a wide range of organisms. In the stimulated PMN, MPO catalyzes the production of hypohalous acids, primarily hypochlorous acid in physiologic situations, and other toxic intermediates that greatly enhance PMN microbicidal activity. Catalytic activity: Cl- + H2O2 + H+ = HClO + H2O. Cofactor: Binds 1 calcium ion per monomer.
