This assay has high sensitivity and excellent specificity for detection of pig PEDF. No significant cross-reactivity or interference between pig PEDF and analogues was observed.

Samples: Serum, plasma and tissue homogenates. Assay Type: Competitive. Detection Range: 0.05 ng/ml-10 ng/ml. Sensitivity: The minimum detectable dose of pig PEDF is typically less than 0.025 ng/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest pig PEDF concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<15%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<15%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for PEDF and Horseradish Peroxidase (HRP) conjugated PEDF. The competitive inhibition reaction is launched between with HRP labeled PEDF and unlabeled PEDF with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of PEDF in the sample. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the serpin family, although it does not display the serine protease inhibitory activity shown by many of the other serpin family members. The encoded protein is secreted and strongly inhibits angiogenesis. In addition, this protein is a neurotrophic factor involved in neuronal differentiation in retinoblastoma cells.[provided by RefSeq, Mar 2011]
