Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for VE-cad has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any VE-cad present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VE-cad is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VE-cad bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene is a classical cadherin from the cadherin superfamily and is located in a six-cadherin cluster in a region on the long arm of chromosome 16 that is involved in loss of heterozygosity events in breast and prostate cancer. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of the cohesion and organization of the intercellular junctions.
