Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for IL-25 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any IL-25 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for IL-25 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of IL-25 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a cytokine that shares sequence similarity with interleukin 17. This cytokine can induce NF-kappaB activation, and stimulate the production of interleukin 8. Both this cytokine and interleukin 17B are ligands for the cytokine receptor IL17BR. Studies of a similar gene in mice suggest that this cytokine may be a pro-inflammatory cytokine favoring the Th2-type immune response. Alternative splicing results in multiple transcript variants.
