Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with Hpt. Standards or samples are then added to the appropriate microtiter plate wells with Horseradish Peroxidase (HRP) -conjugated antibody preparation specific for Hpt, mix well and incubated. The more the amount of Hpt in samples, the less HRP- conjugated antibody preparation specific for Hpt bound by pre-coated Hpt. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. And the color develops in opposite to the amount of Hpt in the sample. The color development is stopped and the intensity of the color is measured.

Haptoglobin captures, and combines with free plasma hemoglobin to allow hepatic recycling of heme iron and to prevent kidney damage. Haptoglobin also acts as an antioxidant, has antibacterial activity and plays a role in modulating many aspects of the acute phase response. Hemoglobin/haptoglobin complexes are rapidely cleared by the macrophage CD163 scavenger receptor expressed on the surface of liver Kupfer cells through an endocytic lysosomal degradation pathway
