Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for CAT has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any CAT present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for CAT is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of CAT bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes catalase, a key antioxidant enzyme in the bodies defense against oxidative stress. Catalase is a heme enzyme that is present in the peroxisome of nearly all aerobic cells. Catalase converts the reactive oxygen species hydrogen peroxide to water and oxygen and thereby mitigates the toxic effects of hydrogen peroxide. Oxidative stress is hypothesized to play a role in the development of many chronic or late-onset diseases such as diabetes, asthma, Alzheimer's disease, systemic lupus erythematosus, rheumatoid arthritis, and cancers.
