Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A antibody specific to the Carrier protein of E2 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with E2 conjugated Carrier protein and Horseradish Peroxidase (HRP) conjugated E2-antibody, then incubated. A competitive inhibition reaction is launched between E2-antibody (Standards or samples) and Horseradish Peroxidase (HRP) conjugated E2-antibody with the E2 conjugated Carrier protein. The more the amount of E2-antibody in samples, the less E2 conjugated Carrier protein bound by Horseradish Peroxidase (HRP) conjugated E2-antibody. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of E2-antibody in the sample. The color development is stopped and the intensity of the color is measured.
