Introduction: Interleukin 6 receptor (IL-6R) also known as CD126 (Cluster of Differentiation 126) is a type I cytokine receptor. IL-6 is a potent pleiotropic cytokine that regulates cell growth and differentiation and plays an important role in immune response. Dysregulated production of IL-6 and this receptor are implicated in the pathogenesis of many diseases, such as multiple myeloma, autoimmune diseases and prostate cancer. In melanocytic cells IL-6R gene expression may be regulated by MITF. The IL-6 receptor is a protein complex consisting of a IL-6 receptor subunit (IL-6R) and interleukin 6 signal transducer Glycoprotein 130. IL-6R also denotes the human gene encoding this subunit. Alternatively spliced transcript variants encoding distinct isoforms have been reported. IL-6R subunit also shared by many other cytokines. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to IL-6R. Standards or samples are then added to the appropriate microtiter plate wells with a biotin conjugated antibody preparation specific for IL-6R and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain IL-6R, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of IL-6R in the samples is then determined by comparing the O.D. of the samples to the standard curve.
