Samples: For the quantitative determination of human Estradiol (E2) concentrations in serum, plasma, cell culture supernates, urine, tissue homogenates. Detection Range: 25pg/ml-650 pg/ml.

Sensitivity: The minimum detectable dose of human E2 is typically less than 25 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest human E2 concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations. Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<15%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<15%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with goat-anti-rabbit antibody. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for E2 and Horseradish Peroxidase (HRP) conjugated E2. The competitive inhibition reaction is launched between with HRP labeled E2 and unlabeled E2 with the antibody. A substrate solution is added to the wells and the color develops in opposite to the amount of E2 in the sample. The color development is stopped and the intensity of the color is measured.
