products description :
Introduction: Matrix metallopeptidase 8 (neutrophil collagenase), also known as MMP8, is a human gene. Proteins of the matrix metalloproteinase (MMP) family are involved in the breakdown of extracellular matrix in normal physiological processes, such as embryonic development, reproduction, and tissue remodeling, as well as in disease processes, such as arthritis and metastasis. Most MMP's are secreted as inactive proproteins which are activated when cleaved by extracellular proteinases. However, the enzyme encoded by this gene is stored in secondary granules within neutrophils and is activated by autolytic cleavage. Its function is degradation of type I, II and III collagens. The gene is part of a cluster of MMP genes which localize to chromosome 11q22.3. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with an antibody specific to MMP-8. Standards or samples are then added to the appropriate microtiter plate wells with a 3 biotin-conjugated antibody preparation specific for MMP-8 and Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. Then a TMB (3,3',5,5' tetramethyl-benzidine) substrate solution is added to each well. Only those wells that contain MMP-8, biotin-conjugated antibody and enzyme-conjugated Avidin will exhibit a change in color. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of MMP-8 in the samples is then determined by comparing the O.D. of the samples to the standard curve.

ncbi summary :
mouse homolog is a neutral metalloproteinase of the fibrillar collagenase family [RGD, Feb 2006]
uniprot summary :
Function: Can degrade fibrillar type I, II, and III collagens. Catalytic activity: Cleavage of interstitial collagens in the triple helical domain. Unlike EC 3.4.24.7, this enzyme cleaves type III collagen more slowly than type I. Cofactor: Binds 3 calcium ions per subunit . Enzyme regulation: Cannot be activated without removal of the activation peptide
