Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for HO-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any HO-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for HO-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of HO-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Heme oxygenase cleaves the heme ring at the alpha methene bridge to form biliverdin. Biliverdin is subsequently converted to bilirubin by biliverdin reductase. Under physiological conditions, the activity of heme oxygenase is highest in the spleen, where senescent erythrocytes are sequestrated and destroyed. Exhibits cytoprotective effects since excess of free heme sensitizes cells to undergo apoptosis. Catalytic activity: Protoheme + 3 AH2 + 3 O2 = biliverdin + Fe2+ + CO + 3 A + 3 H2O.
