Principle of the assay: </b<This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for FXII has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any FXII present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for FXII is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of FXII bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: Factor XII is a serum glycoprotein that participates in the initiation of blood coagulation, fibrinolysis, and the generation of bradykinin and angiotensin. Prekallikrein is cleaved by factor XII to form kallikrein, which then cleaves factor XII first to alpha-factor XIIa and then trypsin cleaves it to beta-factor XIIa. Alpha-factor XIIa activates factor XI to factor XIa . Catalytic activity: Selective cleavage of Arg- -Ile bonds in factor VII to form factor VIIa and factor XI to form factor XIa.
