Introduction: The estrogen-induced protein pS2 is secreted by MCF7 cells upon stimulation by estrogen. Estrogen mediates the transcription of responsive genes via its interaction with the estrogen receptor (ER). ps2 gene is the most interesting predictor for researching the anti-estrogen therapy of breast cancer. But the biological function and role of ps2 is not yet very clear. Now somebody think it has a certain specificity of breast cancer. At the same time, ps2 can reflect the expression of tumor differentiation. Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. A anti-antibody specific to the antibody of pS2 has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with an antibody specific for pS2 and Horseradish Peroxidase (HRP) conjugated pS2, then incubated. A competitive inhibition reaction is launched between pS2 (Standards or samples) and Horseradish Peroxidase (HRP) conjugated pS2 with the antibody specific for pS2. The more the amount of pS2 in samples, the less antibody bound by Horseradish Peroxidase (HRP) conjugated pS2. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of pS2 in the sample. The color development is stopped and the intensity of the color is measured.
