Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for BMP4 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any BMP4 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for BMP4 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of BMP4 bound in the initial step. The color development is stopped and the intensity of the color is measured.

The protein encoded by this gene is a member of the bone morphogenetic protein family which is part of the transforming growth factor-beta superfamily. The superfamily includes large families of growth and differentiation factors. Bone morphogenetic proteins were originally identified by an ability of demineralized bone extract to induce endochondral osteogenesis in vivo in an extraskeletal site. This particular family member plays an important role in the onset of endochondral bone formation in humans, and a reduction in expression has been associated with a variety of bone diseases, including the heritable disorder Fibrodysplasia Ossificans Progressiva.
