Introduction: Dihydrotestosterone (DHT) Full name: 5alpha-Dihydrotestosterone, abbreviating to 5alpha-DHT; INN: androstanolone is a biologically active metabolite of the hormone testosterone, formed primarily in the prostate gland, testes, hair follicles, and adrenal glands by the enzyme 5alpha-reductase by means of reducing the 4,5 double-bond. Dihydrotestosterone belongs to the class of compounds called androgens, also commonly called androgenic hormones or testoids. Androgens are part of the biology of gender by stimulating and controlling the development and maintenance of masculine characteristics. Principle of the Assay: The microtiter plate provided in this kit has been pre-coated with a goat-anti-rabbit antibody. Standards or samples are then added to the appropriate microtiter plate wells with a HRP-conjugated DHT and antibody preparation specific for DHT and incubated. Then substrate solutions are added to each well. The enzyme-substrate reaction is terminated by the addition of a sulphuric acid solution and the color change is measured spectrophotometrically at a wavelength of 450 nm +/- 2 nm. The concentration of DHT in the samples is then determined by comparing the O.D. of the samples to the standard curve.
