Principle of the assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for Apo-A1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any Apo-A1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for Apo-A1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of Apo-A1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes a preproprotein that is proteolytically cleaved to yield a signal peptide and a proproptein that is subsequently processed to generate the active mature peptide. The encoded protein is the major protein component of plasma high density lipoprotein (HDL). This protein facilitates the removal of cholesterol and other fats from tissues by transporting them to the liver for excretion. This protein is a cofactor for lecithin cholesterolacyltransferase, an enzyme that catalyzes the conversion of free cholesterol to cholesteryl esters. Mutations in this gene in humans causes familial HDL deficiency, Tangier disease and familial visceral amyloidosis. Similar clinical features are exhibited by mice with mutations in this gene.
