This assay has high sensitivity and excellent specificity for detection of Bovine cTn-T. No significant cross-reactivity or interference between Bovine cTn-T and analogues was observed.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV% is less than 8%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision (Precision between assays): CV% is less than 10%. Three samples of known concentration were tested in twenty assays to assess. Detection Range: 187.5pg/ml- 3000pg/ml. Detection Wavelength: 450 nm. Sensitivity: 93.75 pg/ml

Principle of the Assay: This assay employs the competitive inhibition enzyme immunoassay technique. An antibody specific to Tn-T has been pre-coated onto a microplate. Standards or samples are added to the appropriate microtiter plate wells with biotin-conjugated Tn-T and incubated. A competitive inhibition reaction is launched between Tn-T (Standards or samples) and Biotin-conjugated Tn-T with the pre-coated antibody specific for Tn-T. The more amount of Tn-T in samples, the less antibody bound by Biotin-conjugated Tn-T. Then Avidin conjugated to Horseradish Peroxidase (HRP) is added to each microplate well and incubated. The substrate solutions are added to the wells, respectively. And the color develops in opposite to the amount of Tn-T in the sample. The color development is stopped and the intensity of the color is measured.

Function: Troponin is the central regulatory protein of striated muscle contraction. Tn consists of three components: Tn-I which is the inhibitor of actomyosin ATPase, Tn-T which contains the binding site for tropomyosin and Tn-C. The binding of calcium to Tn-C abolishes the inhibitory action of Tn on actin filaments.
