This assay has high sensitivity and excellent specificity for detection of sheep TNF-alpha. No significant cross-reactivity or interference between sheep TNF-alpha and analogues was observed.

Unopened test kits should be stored at 2 to 8 degree C upon receipt. Please refer to pdf manual for further storage instructions.

Samples: Serum, plasma, tissue homogenates. Assay Type: Competitive. Detection Range: 3.12 pg/ml -50 pg/ml. Sensitivity: The minimum detectable dose of sheep TNF-alpha is typically less than 3.12 pg/ml. The sensitivity of this assay, or Lower Limit of Detection (LLD) was defined as the lowest sheep TNF-alpha concentration that could be differentiated from zero. It was determined the mean O.D value of 20 replicates of the zero standard added by their three standard deviations.

Intra-assay Precision: Intra-assay Precision (Precision within an assay): CV%<15%. Three samples of known concentration were tested twenty times on one plate to assess. Inter-assay Precision: Inter-assay Precision (Precision between assays): CV%<15%. Three samples of known concentration were tested in twenty assays to assess.

Principle of the assay: This assay employs the competitive inhibition enzyme immunoassay technique. The microtiter plate provided in this kit has been pre-coated with an antibody specific to TNF-alpha. Standards or samples are added to the appropriate microtiter plate wells with Biotin-conjugated TNF-alpha. A competitive inhibition reaction is launched between TNF-alpha (Standards or samples) and Biotin-conjugated TNF-alpha with the pre-coated antibody specific for TNF-alpha. The more amount of TNF-alpha in samples, the less antibody bound by Biotin-conjugated TNF-alpha. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Substrate solution is added to the wells and the color develops in opposite to the amount of TNF-alpha in the sample. The color development is stopped and the intensity of the color is measured.
