Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for EPO has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any EPO present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for EPO is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of EPO bound in the initial step. The color development is stopped and the intensity of the color is measured.

This gene encodes the glycoprotein hormone erythropoietin that regulates the production of red blood cells and biosynthesis of hemoglobin. The predominant expression of this gene shifts from the liver during fetal development to kidney in adults. A complete lack of the encoded protein causes embryonic lethal anemia in mice. The conditional inactivation of this gene in adult mice results in a chronic, normocytic and normochromic anemia. Transgenic mice expressing the human ortholog of this gene exhibit polycythemia.
