Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for SC has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any SC present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for SC is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of SC bound in the initial step. The color development is stopped and the intensity of the color is measured.

Function: The central subunit of the protein translocation channel SecYEG. Consists of two halves formed by TMs 1-5 and 6-10. These two domains form a lateral gate at the front which open onto the bilayer between TMs 2 and 7, and are clamped together by SecE at the back. The channel is closed by both a pore ring composed of hydrophobic SecY resides and a short helix (helix 2A) on the extracellular side of the membrane which forms a plug.
