Principle of the Assay: This assay employs the quantitative sandwich enzyme immunoassay technique. Antibody specific for VEGFR-2/Flk-1 has been pre-coated onto a microplate. Standards and samples are pipetted into the wells and any VEGFR-2/Flk-1 present is bound by the immobilized antibody. After removing any unbound substances, a biotin-conjugated antibody specific for VEGFR-2/Flk-1 is added to the wells. After washing, avidin conjugated Horseradish Peroxidase (HRP) is added to the wells. Following a wash to remove any unbound avidin-enzyme reagent, a substrate solution is added to the wells and color develops in proportion to the amount of VEGFR-2/Flk-1 bound in the initial step. The color development is stopped and the intensity of the color is measured.

Vascular endothelial growth factor (VEGF) is a major growth factor for endothelial cells. This gene encodes one of the two receptors of the VEGF. This receptor, known as kinase insert domain receptor, is a type III receptor tyrosine kinase. It functions as the main mediator of VEGF-induced endothelial proliferation, survival, migration, tubular morphogenesis and sprouting. The signalling and trafficking of this receptor are regulated by multiple factors, including Rab GTPase, P2Y purine nucleotide receptor, integrin alphaVbeta3, T-cell protein tyrosine phosphatase, etc. Mutations of this gene are implicated in infantile capillary hemangiomas.
